Summary
The protein VE-cadherin is expressed selectively by all endothelial cells and is an important regulator of endothelial permeability. Traditional methods of visualizing endothelial junctions have relied on fixed tissue antibody staining or exogenous labeling methods, which often result in variable staining efficiency, inability to capture dynamics, or disruption of native protein levels.
Our researchers developed a VE-cadherin reporter mouse model of where the red fluorescent protein mScarlet is added at the 3’ end of the endogenous Cdh5 (VE-cadherin) locus that yields expression under the native VE-cadherin promoter in all endothelial cells at physiological levels, enabling precise, physiologically relevant visualization of VE-cadherin live tissue dynamics. It offers a powerful, non-invasive tool that uniquely facilitates live, in vivo investigation of endothelial junction dynamics.
